Evaluation of the effect of Plant Growth Regulators and salinity on in vitro micropropagation of Salicornia iranica Akhani populations

Abstract

A large part of Iran is in a state of salinity and semi-salinity. In such areas, the best solution is to use plants that can tolerate salinity. Salicornia halophyte plant its multiple uses and Micropropagation of Salicornia plays important role the management of saline areas. In the present study, the seeds of three populations of Salicornia were collected from around of Lake Urmia, after preparing the seedlings in the greenhouse, they were used for shooting with different concentrations of growth regulators (6- Benzylaminopurine and 1- Naphthaleneacetic acid) and sodium chloride and also for rooting, different culture mediums were studied in completely randomized factorial design with three replications. The results showed that there is a significant difference between different levels of BAP, NAA, NaCl and Salicornia populations in shoot length trait and number of nodes. Based on the comparison of means, the population of Dizaj-e Dowl in combination with 400 mM chloride-sodium and also the combination of 400 mM chloride-sodium with each of the three levels of BAP caused the highest shoot length. The same population in combination with 400 mM sodium chloride and 3 mg/L BAP and treatment combinations containing 0.5 and 1 mg/L NAA and 3 mg/L BAP had the highest number of nodes. For rooting traits, there was also a significant difference between different rooting media. Dizaj-e Dowl population also had the highest number of roots in combination with M5 medium (M5P3). In other words, Dizaj-e Dowl population was introduced as the best population in all studied traits.
