چکیده
رشد گیاهان به شدت تحت تأثیر تنشهای محیطی چون، خشکی، شوری زیاد، درجه حرارت کم یا زیاد قرار میگیرد و بر این اساس شناسایی ژنها، بهخصوص ژنهای تنظیمکننده که در انطباق یا تحمل تنش نقش دارند بسیار ضروری است. پروتئینهای خانواده MYB یکی از خانوادههای بزرگ عوامل نسخه برداری اختصاصی گیاهی است و نقشهای متنوعی را در مراحل نمو گیاهی و پاسخ به تنشها ایفا میکند. از این رو هدف از این تحقیق بررسی الگوی بیان نسبی ژن TaMYB73 و همچنین تغییرات محتوی تنظیمکنندههای اسمزی، محتوی آب نسبی برگ، میزان پروتئین و رنگیزههای فتوسنزی چهار رقم جو منطقه سیستان (نیمروز، نومار،اکلیل و زهک) تحت سطوح مختلف تنش خشکی (100 درصد ظرفیت زراعی (شاهد)، 75 درصد ظرفیت زراعی و 50 درصد ظرفیت زراعی) بود. این آزمایش به صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار انجام شد. نتایج مقایسه میانگین نشان داد با افزایش سطوح تنش خشکی نسبت به سطح شاهد، میزان رنگیزههای فتوسنتزی، محتوی نسبی آب برگ و پروتئین کاهش یافت ولی بیان نسبی ژن ژنTaMYB73 ، غلظت پرولین، کربوهیدرات و میزان کاروتنوئید افـزایش یافت. در بین ارقام مورد بررسی میزان تغییرات تنظیمکنندههای اسمزی، رنگدانههای فتوسنتزی، محتوی آب نسبی برگ و بیان نسبی ژن مورد مطالعه در رقم نیمروز و سپس اکلیل، نسبت به سایر ارقام بالاتر بود. از این رو پیشنهاد میشود در مطالعات بعدی سایر آزمایشات تکمیلی نیز برای تأیید نتایج فوق انجام شود تا بتوان از طریق مطالعات آزمایشگاهی ارقام را از نظر تنشها و بهخصوص تنش خشکی در زمان کمتری دستهبندی نمود.

Investigation of relative expression of TaMYB73 gene in some native barley of Sistan region under drought stress

Plant growth is greatly influenced by environmental stresses including water deficit, high salinity and extreme temperatures. Therefore, the identification of genes especially those regulatory ones whose expression enables plants to adapt to or to tolerate these stresses, is essential. MYB proteins belong to a big family of transcription factors of plants and they are also active in different stages of plants’ growth and their response to stress. This study was aimed at investigating the expression pattern of TaMYB73 gene, changes in the osmotic regulators content, relative water content in leaves, protein as well as photosynthetic pigments contents of four barley cultivars native to Sistan region (Aklil, Nomar, Nimroz, Zahak) under different levels of drought stress (100% (control), 75% and 50% field capacity). This experiment was performed as a factorial in a completely randomized design with three replications. Results indicated that increasing drought stress levels led to a decrease in the amount of photosynthetic pigments, protein and relative leaf water contents. However, proline and carbohydrate concentrations, carotenoid content and the relative expression of TaMYB73 gene increased. Among the cultivars investigated in this research, the rate of changes in osmotic regulators content, photosynthetic pigments and relative water contents as well as relative expression of TaMYB73 gene in Nimroz cultivar was the highest followed by Aklil cultivar. In order to facilitate the classification of resistant cultivars in terms of stresses, drought stress in particular, through laboratory studies, it is suggested that additional tests be performed in future investigations to confirm the above results.
