Complex eukaryotic organisms evolved in response to developmental and environmental signs such as UV-B radiation through the selective expression of a specific section of the same genome in different cell types. The foundation of multicellular organisms needs genetically predefined signaling pathways in diverse cell types. Cell types should be enabled to modulate appropriate adaptive reactions in ever-changing comprehensive environment. The multicellular green alga Volvox carteri consists of only two types with a distinct division of function. Here, the RNA-seq technique was employed to investigate transcriptome profiling of two separated cell types of V. carteri under 1h UV-B treatment. Our results revealed that almost one-third of the V. carteri genes (8392 genes out of 23778 genes) were significantly differentially expressed between the two cell types. The results showed that in somatic cells, potassium ion transmembrane transport and RNA phosphodiester bond hydrolysis were activated. However, in reproductive cells, mismatch repair (MMR), nucleosome assembly and chromosome assembly occurred after UV-B radiation. RNA-dependent DNA biosynthetic process was a common response of both cells to UV-B radiation. The cell type-specific genes were examined to determine cell function in response to UV-B radiation. Overall, two cell-types have initiated defense mechanisms and decreased the sensitivity of alga to UV-B radiation.}, keywords = {Volvox carteri,UV-B,Transcriptomics,RNA-seq,Reproductive and somatic cells}, title_fa = {بررسی الگوی بیان ژن ویژه تیپ سلولی در جلبک Volvox carteri تحت شرایط تنش نوری}, abstract_fa = {طول موج‌های UV-B عمیق‌تر از هر طول موج دیگری به آب نفوذ می‌کنند. موجودات یوکاریوتی پیچیده از طریق بیان انتخابی بخش ویژه‌ای از ژنوم یکسان در انواع مختلف سلول، در پاسخ به نشانه‌های نموی و محیطی از قبیلUV-B تکامل یافته‌اند. تشکیل موجودات پرسلولی به مسیر‌های سیگنالینگ ژنتیکی از پیش تعریف شده در انواع مختلف سلول نیاز دارد. انواع مختلف سلول باید بتوانند پاسخ‌های سازشی مناسب را نسبت به محیط در حال تغییر پیرامون خود، تنظیم کنند. جلبک سبز پرسلولیVolvox carteri تنها از دو تیپ سلولی با تقسیم بارز کار تشکیل شده است.در اینجا، از تکنیکRNA-seq برای بررسی پروفایل ترنسکریپتوم دو تیپ سلولی تفکیک شده V. carteri تحت یک ساعت تابش UV-B استفاده شد. نتایج نشان داد که تقریبا یک سوم از ژن‌ها (8392 ژن از 23778 ژن) به‌طور معنی‌داری بین دو تیپ سلولی بیان متفاوتی داشته‌اند. برای شناسایی نحوه عملکرد‌ هر یک از سلول‌ها در پاسخ به تابش UV-B، ژن‌های اختصاصی آنها مورد بررسی قرار گرفت. نتایج نشان داد که در سلول‌های سوماتیک، انتقال تراغشایی پتاسیم و هیدرولیز باند فسفودی‌استراز RNA فعال شده‌اند. درحالی‌که در سلول‌های زایشی، ترمیم برشی عدم تطابق (MMR)، گردایش نوکلئوزوم و کروموزوم بعد از تابش UV-B رخ‌داده‌است. فرایند بیوسنتزDNA وابسته به RNA، پاسخ مشترک هر دو سلول به تابش UV-B بوده است. هر دو تیپ سلولی در مجموع سازوکار‌های دفاعی را آغاز‌کرده‌اند و باعث کاهش حساسیت جلبک به این اشعه شده‌اند.}, keywords_fa = {Volvox carteri,UV-B,Transcriptomics,RNA-seq,Reproductive and somatic cells}, url = {https://plant.ijbio.ir/article_1631.html}, eprint = {https://plant.ijbio.ir/article_1631_5dc87a57da9c2b52b31db6703ad07b7e.pdf} }
