بررسی کالزایی و باززایی در گونه جنگلی آزاد (Zelkova carpinifolia) در شرایط کشت درون شیشهای
درخت آزاد (Zelkova carpinifolia)، یکی از درختان جنگلی در معرض تهدید در ایران و جهان است که در حال حاضر به بیماری جهانی مرگ نارون توسط قارچ Ophiostoma novo-ulmi نیز مبتلا شده است. بطوریکه ذخیرهگاههای جنگلی این گونه در اثر ابتلا به این بیماری با سرعت بالایی در حال از بین رفتن هستند. این تحقیق به منظور بررسی کالزایی و بازایی در گونه جنگلی Z. carpinifolia در شرایط کشت درون شیشه انجام گرفت. آزمایشهای بررسی تاثیر محیط کشت و ترکیبات هورمونی، ژنوتیپ و اثرات متقابل آنها بر درصد کالوسزایی و جوانهزنی کالوسها در قالب طرح کاملا تصادفی با دو نوع ریزنمونه برگ و ساقه انجام شدند. نتایج ریزازدیادی نشان داد که درصد کالوسزایی در برگ (2/82 درصد)> نسبت به ریزنمونه ساقه (9/72 درصد) بیشتر بود. محیط کشت MS حاوی 2 میلیگرم در لیتر BAPو 1 میلیگرم در لیتر NAA بالاترین میزان-کالوسزایی (3/94 درصد) را داشت و به عنوان بهترین محیط کشت در نظر گرفته شد. بیشترین درصد القای تشکیل جوانه از کالوس (97/7 درصد) بر روی محیط کشت MS به همراه 05/0 میلیگرم در لیتر GA3 مشاهده شد. محیط کشت MS غنی شده با IBA (2 میلیگرم بر لیتر) و NAA (2 میلیگرم بر لیتر) با 70 درصد ریشهدهی، بهترین محیط ریشهزایی بود.

بررسی مقاومت به خشکی چند رقم انگور در شرایط آزمایشگاهی
در این مطالعه اثر غلظتهای مختلف پلیاتیلنگلیکول و سوربیتول بر روی ارقام حسینی، قره ملیحی، دست-ارچین، اینهامجیی، سیاه، ریشبابا، طولاگوزو و رشه انگور بررسی شد و صفات تعداد و طول ساقه، تعداد برگ، تعداد و طول ریشه، تعداد و طول ریشههای فرعی اندازهگیری شدند. نتایج تجزیه واریانس دادهها نشان داد اثر ارقام بر روی صفات طول ساقه، تعداد برگ، طول ریشه و صفت تعداد ریشههای فرعی و اثر غلظتهای مختلف پلیاتیلنگلیکول بر تمامی صفات در سطح احتمال 5% معنیدار بود. نتایج نشان داد شاخصهای رشدی در غلظت 60 گرم بر لیتر کمترین رشد و در تیمار شاهد بیشترین افزایش مشاهده گردید. مقایسه میانگین دادهها مربوط به فاکتور ارقام نشان داد که به ترتیب بیشترین و کمترین مقدار طول ریشه٬ تعداد برگ و تعداد ریشههای فرعی را ارقام طولاگوزو و اینهامجیی تولید نمودند. نتایج تجزیه واریانس دادههای مربوط به سوربیتول نشان داد اثر ارقام و سوربیتول بر روی تمام صفات مورد بررسی در سطح احتمال 5% معنیدار بودند. تیمار شاهد و غلظت 30 گرم در لیتر سوربیتول به ترتیب کمترین و بیشترین مقدار را در صفات تعداد و طول ساقه، تعداد برگ و تعداد و طول ریشه را داشتند. مقایسه میانگین سوربیتول نشان داد که به ترتیب بیشترین و کمترین مقدار را ارقام سیاه و اینهامجیی تولید نمودند.

بررسی تاثیر تنظیم کنندههای رشد گیاهی بر روی شاخهزایی و ریشهزایی گلمحمدی Rosa damascena Mil.)l.) بومی آذربایجان شرقی در شرایط درون شیشهای
گلمحمدی (.Rosa damascena Mill.) یکی از مهمترین رزهای معطر است که به منظور تولید گلاب، اسانس و مصارف دارویی کشت میگردد. این پژوهش به منظور بررسی اثر تنظیم کنندهای رشد گیاهی بر روی قدرت شاخهزایی، ریشهزایی و سازگاری در شرایط گلخانه انجام شد. به منظور سترونسازی ریزنمونهها از هیپوکلریت سدیم 10 درصد به مدت 15 دقیقه و الکل 70 درصد استفاده گردید. تیمارهای استقرار شامل هورمون سایتوکنین BA و BAP در سه سطح (0، 5/0 و 1)، تیمارهای شاخهزایی BAP و BA در چهار سطح (0، 1، 2 و 3) و تیمارهای ریشهزایی با هورمونIBA در سه سطح (0، 5/0 و 1) میلی گرم در لیتر مورد بررسی قرار گرفتند. کلیه تیمارها در محیط کشت MS در سه تکرار و هر تکرار با 3 ریزنمونه در قالب طرح کاملا تصادفی کشت گردید. تجزیه دادهها نشان داد که بیشترین میزان استقرار 88/89 درصد در 0/5 میلی گرم در لیتر BA و کمترین میزان استقرار 25 درصد در تیمار صفر BAP حاصل شد. مقایسه میانگین داده ها نشان داد که بیشترین ضریب شاخهزایی 3/38 ، بیشترین طول شاخه 5/34cm در تیمار حاوی 3 میلی گرم در لیتر BA و کمترین ضریب شاخهزایی 1/00 در تیمار بدون هورمون حاصل شد. بیشترین مقدار ریشه زایی 85 درصد در محیط کشت 1/2MS حاوی 0/5 میلی گرم در لیتر IBA حاصل شد و کمترین میزان ریشهزایی 36 درصد بود که در محیط کشت 1/2MSبدون هورمون حاصل شد.

معرفی فلور، شکل زیستی و انتشار جغرافیایی عناصر گیاهی منطقه حفاظت شده قمصر در استان اصفهان
رستنیهای هر منطقه نتیجه واکنشهای جامعه زیستی در برابر شرایط محیطی آن است. هدف پژوهش حاضر معرفی فلور، شکل زیستی و گونههای در معرض خطر منطقه حفاظت شده قمصر است. این منطقه با مساحت تقریبی 36700 هکتار در 35 کیلومتری جنوب غربی کاشان قرار دارد. نتایج نشان داد، مجموعه رستنیهای این منطقه، بالغ بر 576 گونه گیاهی متعلق به 80 تیره و 355 جنس است. از مجموع 355 جنس، 50 جنس (7/8 درصد) و 69 گونه (9/11 درصد) به رده تک لپهایها و 299 جنس (9/51 درصد) و 500 گونه (8/86 درصد) به رده دولپهایها تعلق دارد. بزرگترین تیرههای گیاهی از لحاظ تعداد گونه عبارتند از: Compositaeبا 76 گونه،Labiatae وGraminae هر کدام با 50 گونه، Fabaceae با 47 گونه، Cruciferae با 37 گونه، Umbelliferae با 35 گونه و Caryophyllaceae با 28 گونه. بررسی انتشار جغرافیائی رستنیهای منطقه نشان داد، بیشترین انتشار جغرافیائی به ناحیه رویشی ایرانی-تورانی با 381 گونه (1/66 درصد) و ایرانی-تورانی/اروپا-سیبری/مدیترانهای با 33 گونه (7/5 درصد) مربوط است که از بین آنها 18 گونه انحصاری ایران (1/3 درصد) بودند. در بین گیاهان منطقه، همیکریپتوفیتها با 2/32 درصد و تروفیتها با 9/31 درصد، مهمترین شکلهای زیستی را تشکیل دادند. بر اساس معیارهای IUCN و کتابRed Data Book of Iran ، 33 گونه تهدید شده وجود دارد که همگی متعلق به منطقه ایرانی-تورانی است. از این 33 گونه، 2 گونه در معرض خطر، سه گونه در طبقه آسیبپذیر، 26 گونه در طبقه کمتر در معرض خطر و 2 گونه در طبقه کمبود داده قرار دارند.

بررسی تشریحی ساقه و برگ جنس برگ سنایی ( Thalictrum) از تیره آلاله در ایران
گیاه برگ سنایی (L. Thalictrum) یکی از جنس های بزرگ تیره آلاله (Ranunculaceae) است. در این بررسی 16 جمعیت متعلق به شش گونه و دو واریته از این جنس (Th. mazandaranicum، Th. minus var. majus و Th. sultanabadense و Th. isopyroides ,Th. foetidum Th. tacabicu ) مورد مطالعه قرار گرفتند نمونه ها ی بررسی شده از سراسر نواحی پراکنش شان جمع آوری شده و پس از برش گیری دستی و رنگ آمیزی مضاعف مطالعه شدند. صفات تشریحی ساقه مانند: شکل برش عرضی ساقه، تعداد لایه های کلانشیم، تعداد لایه های فیبر اسکلرانشیم، تعداد لایه های سلول های آبکش داخلی، حضور و تعداد لایه های پارانشیم محوری، تعداد حلقه های دستجات آوندی و صفات تشریحی اپیدرم برگ مانند: تعداد سلول های همراه، شکل سلول های اپیدرمی، تیپ روزنه ای، طول سلول های روزنه، کرک های سطح اپیدرم مهمترین صفات در جداسازی گونه های برگ سنایی بودند. اختلاف فاحش در اپیدرم برگی گونههای این جنس، راهکاری مطمئن جهت جداسازی گونههای این جنس از یکدیگر می-باشد. در مجموع تعداد لایه های پارانشیم نردبانی، وجود سلول های فیبر اسکلرانشیم در اطراف دستجات آوندی در رگبرگ میانی، وجود سلول های کلانشیم و انواع کرک،مهمترین صفات تشریحی برگ ها بودند که می توانستند گونه ها را از یکدیگر مشخص نمایند.

اثر محلولپاشی برگی سالیسیلیکاسید و سولفاتپتاسیم بر کمیت و کیفیت اسانس شمعدانی عطری (Pelargonium graveolens)
شمعدانیعطری (Pelargonium graveolens) یک گیاه گلدانی زینتی برگسارهای است که استفاده گستردهای در منازل دارد. افزون بر این به دلیل داشتن خواص دارویی ارزشمند، به طور روزافزون در صنایع عطرسازی و تولید مواد آرایشی و دارو کاربرد دارد. بر این اساس به منظور بهبود ویژگیهای بیوشیمیایی این گیاه زینتی آپارتمانی، آزمایشی به صورت فاکتوریل بر پایه طرح کامل تصادفی با 4 تکرار در گلخانه بخش علوم باغبانی دانشگاه شیراز اجرا شد. محلولپاشی برگی سالیسیلیکاسید و سولفاتپتاسیم در 4 غلظت به ترتیب 0، 100، 200 و 300 میلیگرم بر لیتر، و 0، 5/0، 1 و 5/1 درصد انجام شد. نتایج نشان داد که برهمکنش سالیسیلیکاسید و سولفاتپتاسیم بر کمیت و کیفیت ماده مؤثره برگ شمعدانی عطری اثر داشت. با توجه به نتایج بهدست آمده تیمار 200 میلیگرم بر لیتر سالیسیلیکاسید و 5/1 درصد سولفاتپتاسیم بیشترین عملکرد اسانس (01/ 1 درصد) را به همراه داشت. بیشترین مقدار ترکیبات ژرانیول و سیترونلول در تیمار 300 میلی-گرم بر لیتر سالیسیلیکاسید و شاهد سولفات پتاسیم حاصل شد.

اثرات برهمکنش شوری و منیزیم روی روابط آبی و یونی اسفندک ( Zygophylum fabago.L)
به منظور بررسی اثر متقابل شوری و منیزیم بر رشد، ویژگیهای فیزیولوژیک و محتوای برخی عناصر غذایی در گیاه اسفندک، آزمایشی به صورت فاکتوریل طراحی و در پرلیت با استفاده از محیط کشت هوگلند اجرا شد. تیمارها شامل دو سطح شوری (صفر و300 میلیمولار) و سه سطح منیزیم صفر و 2 و 6 میلیمولار مازاد بر مقدار منیزیم موجود در محلول هوگلند (2 میلیمولار) بودند. کاربرد منیزیم در شرایط شور منجر به افزایش وزن خشک بخشهوایی به بیش از دو برابر تیمار شاهد (55 گرم وزن خشک) و ریشه بیش از 30% شد. شوری موجب افزایش محتوای آب در واحد سطح برگ گردید. در حالیکه تاثیر شوری و منیزیم منجر به کاهش نسبی آن تا حدود تیمار شاهد (400گرم آب برمترمربع) شد. شوری و بر همکنش آن با منیزیم منجربه منفیتر شدن پتانسیل اسمزی برگها شد، که سهم سدیم در این منفیتر شدن در تیمارهای شوری بیشتر از شاهد بود ولی برهمکنش شوری و منیزیم بهشدت موجب کاهش سهم پتاسیم در پتانسیل اسمزی گردید. شوری و منیزیم موجب کاهش شدید کلسیم، منیزیم و پتاسیم و افزایش شدید در محتوای سدیم اندامهای اسفندک شد. بهنظر میرسد که افزودن مقدار منیزیم مناسب به محیط رشد اسفندک، میتواند از طریق ایجاد تغییرات مناسب در نحوه جذب و انباشتگی سایر عناصر، رشد گیاه را در شرایط شور بهبود ببخشد. همچنین غلظت بالای سدیم در برگهای اسفندک به طور مستقیم برای تنظیم اسمزی استفاده میشود که منجر به کاهش پتانسیل آب برگ، بهبود وضعیت آبی برگ و از این طریق بهبود رشد میشود.

ریخت شناسی، تشریح و تنوع ریختی گونه Hypnea pannosa در سواحل خلیج فارس و دریای عمان
هیپنهآ (Hypnea Lamouroux) از جلبکهای قرمزی است که به دلیل تولید کاراژینان در بسیاری از کشورها کشت میشود. گونههای مختلف این جنس در مناطق زیر جزرومدی آبهای گرم حضور دارند و این جنس شامل حدود 53 گونه در سراسر جهان است که در ایران حدود 9 گونه از این جلبک گزارش شده است. گونههای مختلف این جنس روی صخرهها، سنگها، صدفها یا بهصورت اپیفیت با سایر جلبکها رشد میکنند. در این مطالعه ریختشناسی، تشریح و تنوع ریختی گونهٔ J. Agardh H. pannosa برای اولین بار در ایران انجام شده است. در این مطالعه، 15 فرد از 7 جمعیت بررسی شد. تعداد 13 صفت کمی و کیفی اندازهگیری و استاندارد شد و در تحلیلهای آماری استفاده شد.تفاوتهایی در ریختشناسی جمعیتهای مورد مطالعه مشاهده شد بهطوریکه جمعیتهای نمونهبرداری شده از دریای عمان (تیس و رمین) توسط صفات رنگ، فراوانی انشعابات جانبی و تعداد لایههای سلولهای مدولاری از جمعیتهای نمونهبرداری شده از خلیجفارس (قشم و بندرلنگه) جدا شدند.

تاثیر تیمار قبل و پس از برداشت گاما آمینو بوتیریک اسید (GABA) بر کنترل سرمازدگی گل شاخه بریده آنتوریوم (.Anthurium andraeanum L)
به دلیل حساسیت گلهای شاخه بریده آنتوریوم به سرمازدگی، دمای بهینه نگهداری آنها 5/12 تا 20 درجه سانتیگراد میباشد. در این پژوهش، تاثیر تیمار گاما آمینوبوتیریک اسید (GABA) در غلظتهای صفر (شاهد)، 1، 5، 10، 15 و 20 میلی مولار به صورت اسپری قبل از برداشت و غوطهوری انتهای ساقه پس از برداشت (15 دقیقه در دمای 20 درجه سانتیگراد) بر سرمازدگی پس از برداشت گل شاخه بریده آنتوریوم رقم سیریون نگهداری شده در دمای 4 درجه سانتیگراد به مدت 21 روز مورد ارزیابی قرار گرفت. سرمازدگی در گل شاخه بریده آنتوریوم با قهوهای شدن اسپات به همراه افزایش نشت یونی و تجمع مالون دی آلدئید (MDA) همراه بود. تیمار GABA در غلظتهای 1 و 5 میلی مولار، به ترتیب قبل و پس از برداشت، موجب کاهش قهوهای شدن اسپات گردید و افزایش نشت یونی و تجمع MDA را به تاخیر انداخت. تیمار GABA کاهش محتوای نسبی آب (RWC) در اسپات گل های شاخه بریده آنتوریوم در طول 21 روز نگهداری در دمای 4 درجه سانتی گراد را به تاخیر انداخت. علاوه بر این، در طول 21 روز نگهداری در دمای 4 درجه سانتی گراد، تیمار GABA کاهش آنتوسیانین را کندتر نموده و گلهای شاخه بریده آنتوریوم تیمارشده با GABA دارای سطوح بالای آنتوسیانین بودند. تیمار GABA موجب افزایش تجمع گلایسین بتائین (GB) در گلهای شاخه بریده آنتوریوم در دمای 4 درجه سانتیگراد گردید.

اثرات تغذیه سیلیکون بر کاهش تنش اکسیداتیو ناشی از شوری در گیاه آرابیدوپسیس تالیانا (Arabidopsis thaliana)
در این پژوهش اثرات تغذیه سیلیکون در گیاه آرابیدوپسیس تحت تنش شوری در شرایط کشت هیدروپونیک مورد مطالعه قرار گرفت. آزمایشات در قالب طرح کاملاً تصادفی و به صورت فاکتوریل انجام شد. فاکتور اول، شوری در سطوح 0، 50، 100 و 150 میلیمولار به صورت کلرید سدیم و فاکتور دوم، سیلیکون در سطوح 0 و 5/1 میلیمولار به صورت سیلیکات سدیم بود. نتایج کشت گیاهان آشکار ساخت که در نتیجه تنش شوری تا تیمار 150 میلیمولار میزان یون سدیم در گیاه افزایش و درصد آب نسبی و مقدار کلسیم، پتاسیم و منیزیم به ترتیب در حدود 27، 49، 66 و 19 درصد نسبت به گیاه شاهد کاهش یافته است. این امر احتمالاً با القا تنش اکسیداتیو سبب افزایش فعالیت آنزیمهای آنتیاکسیدان و مقدار فنلها شد که منجر به کاهش رشد گیاهان تحت تنش شوری گردید. به علاوه، تنش شوری قندهای محلول را افزایش و نشاسته را کاهش داد که احتمالاً تلاش گیاه برای تنظیم اسمزی ناکافی را آشکار میسازد. تغذیه سیلیکون همراه با افزایش مقدار پتاسیم و منیزیم و نیز افزایش فعالیت آنزیمهای آنتیاکسیدان، کاهش تنش اکسیداتیو ناشی از شوری را سبب شد. به علاوه، افزایش مقدار قندهای احیایی، کاهش نشاسته و افزایش میزان آب نسبی با تغذیه سیلیکون در گیاهان تحت شوری بهبود وضعیت آب گیاهان را نشان میدهد. در نتیجه گیاهان تغذیهشده با سیلیکون تحت شوری نسبت به گیاهان بدون سیلیکون رشد بهتری داشتند. این نتایج نشان داد که سیلیکون با کاهش تنش اکسیداتیو و بهبود وضعیت آب، تنش شوری را در گیاه آرابیدوپسیس تخفیف میدهد.

بررسی تأثیر تنظیم کنندههای رشد بر ریزازدیادی درونشیشهای بنفشه آفریقایی (,Saintpaulia, Pretty Miss Kelly) از ریزقطعات برگ
استفاده از فناوری کشت بافت میتواند از جمله مطلوبترین روشهای تکثیر سریع گیاهان زینتی در زمانی کوتاه و فضایی محدود محسوب می شود. این پروژه به منظور دستیابی به مناسبترین محیط غذایی جهت تکثیرانبوه گیاه زینتی بنفشه آفریقایی از ریزقطعات برگ در شرایط درونشیشهای انجام شد. اثر تنظیمکنندههای مختلف بر میزان تولید شاخساره، تعداد برگ، تعداد ریشه و مدت زمان ریشهزایی بررسی شد. لذا برگهای جوان از پایه مادری رقم 'Pretty Miss Kelly' جدا و پس از ضدعفونی به ریزقطعات با اندازه یک سانتیمترمربع تقسیم شدند. اثر تنظیمکننده-های رشدی شامل BAP به همراه NAA یا IAA بصورت ترکیبی و در سطوح مختلف بر تولید شاخساره، اثر BAP و AdS بر تعداد برگ ، اثر محیطهای مختلف ریشه زایی مورد بررسی قرار گرفت. جهت آنالیز دادهها از آزمایش فاکتوریل بر مبنای طرح کاملاً تصادفی استفاده شد. نتایج این بررسی نشان داد که mg/l NAA 0.1 یا mg/l NAA0 به همراه mg/l0.1 از BAP و همچنین mg/l IAA0 به همراه BAP mg/l0.1 مناسبترین ترکیب برای تولید شاخساره هستند. افزایش تعداد برگ بترتیب با استفاده از AdS mg/l 15 و BAP mg/l 0.2 رخ داد. ماسه استریل با تولید میانگین حدود 25 ریشه به در هر گیاهچه نسبت به دو محیط دیگر مناسبتر بود. در زمینه زمان ریشهدهی محیط MS 2/1 به همراه 0.005 میلیگرم در لیتر NAA با میانگین 10 روز مناسب ترین محیط بود. بنابراین در این مطالعه مناسبترین محیط های غذایی در مراحل مختلف جهت تکثیر انبوه بنفشه آفریقایی معرفی شد.

بررسی محتوای عناصر ماکرو،میکرو، میزان کلروز و شاخص کلروفیل ارقام انتخابی بادام پیوند شده روی پایه GF677 در شرایط قلیایی
در این تحقیق واکنش ارقام انتخابی بادام شامل تونو، نانپاریل، مامایی، شکوفه، سهند، آذر، شاهرود 12، A200، A230 و 7-9 پیوند شده روی پایه و خود پایه GF677 در قالب آزمایش فاکتوریل بر پایه طرح کاملاً تصادفی با سه تکرار مطالعه شد. نتایج نشان داد، با افزایش غلظت بیکربنات پتاسیم در آب آبیاری، درصد نیتروژن، پتاسیم، کلسیم، منیزیم و فسفر و محتوی روی و آهن کاهش و درصد نکروزه شدن برگ ها، افزایش یافت ولی میزان افزایش و یا کاهش در صفات اندازه گیری شده در بین رقم های پیوندی با یکدیگر، اختلاف معنی داری را نشان داد. در مجموع کل صفات اندازه گیری شده، رقم شاهرود 12 و پس از آن رقم های A230 و شکوفه به عنوان متحمل ترین و ژنوتیپ 7-9 و رقم سهند به عنوان حساس ترین ارقام نسبت به غلظت های بالای بیکربنات پتاسیم تشخیص داده شدند. رقم های شاهرود 12، A230 و شکوفه به خوبی توانستند غلظت 15 میلی مولار بیکربنات پتاسیم در آب آبیاری را تحمل کنند. همچنین رقم شاهرود 12 تا حدی توانست غلظت 30 میلی مولار بیکربنات پتاسیم در آب آبیاری را نیز، تحمل نماید. همچنین نتایج حاصل از این تحقیق نشان داد که نوع رقم پیوندی در افزایش مقاوت پایه های GF677 به غلظت های بالای بیکربنات پتاسیم موثر است و اغلب ارقام پیوندی (به جز ژنوتیپ 7-9 و رقم سهند)، دارای مقاومت بیشتری به غلظت های بالای بیکربنات پتاسیم در مقایسه با پایه های غیر پیوندی بودند.

اثر مایه زنی توام قارچ اندومیکوریز و باکتریهای ریزوبیوم ملیلوتی و سودوموناس آئروژینوزا برگیاه یونجه (Medicago sativa) در شرایط تنش آبی
چکیدهگیاهان در معرض انواع تنشهای زیستی و غیر زیستی از جمله خشکی قرار می گیرند که بر رشد و محصول دهی آنها تاثیر منفی دارد. همزیستی گیاهان با قارچ میکوریزی از رایج ترین استراتژیهای گیاهی به منظور کنار آمدن با تنشهای محیطی می باشد. به همین منظور در پژوهش حاضر برهمکنش قارچ اندومیکوریز و باکتریهای سودوموناس آئروژینوزا و ریزوبیوم ملیلوتی در آزمایشی به صورت فاکتوریل بر پایه بلوکهای کاملا تصادفی در سه سطح آبیاری و سه تکرار بر گیاه یونجه مورد بررسی قرار گرفت. اسپور قارچ اندومیکوریز همزیست با یونجه از منطقه باقرآباد کرمان در مجاورت گیاه ذرت تکثیر و مورد استفاده قرار گرفت. بذر یونجه رقم بمی با اسپور قارچ مزبور و باکتریها به صورت منفرد و توام تلقیح شدند و در سه سطح آبیاری 70، 50 و 20 درصد ظرفیت زراعی بمدت سه ماه پرورش داده شدند. نتایج نشان داد تنش آبی به طور معنی دار باعث کاهش پروتئین ولی افزایش پرولین وقندهای احیائی در گیاه یونجه شد اما نیتروژن تغییر معنی داری نداشت. تلقیح با قارچ اندومیکوریز و باکتریها میزان پروتئین، پرولین، قندهای احیائی و نیتروژن را در شرایط تنش و غیر تنش در مقایسه با شاهد به طور معنی داری افزایش داد. علاوه بر این اندومیکوریز و باکتریها بر یکدیگر اثر هم افزایی نشان دادند به طوری که در اکثر موارد تلقیح توام موثرتر از تلقیح منفرد بود. بر اساس این نتایج تلقیح توام باکتریهای سودوموناس آئروژینوزا، ریزوبیوم ملیلوتی و قارچ اندومیکوریز به منظور افزایش تحمل گیاه یونجه در برابر تنش آبی پیشنهاد می گردد.

تحلیل ساختار تیپ های مختلف در جنگل های ناو اسالم گیلان با استفاده از شاخص های نزدیک ترین همسایه
توصیف کمی ساختار جنگل به عنوان یکی از مناسب ترین ابزارها در مدیریت نوین جنگل در نظر گرفته می شود. در این تحقیق ساختار گونه های درختی تیپ های راش (Fagus)، راش- ممرز (Fagus-Carpinus) و راش- افرا (Fagus-Acer) جنگل های هیرکانی مورد بررسی و مقایسه قرار گرفت. به منظور انجام این تحقیق 5 قطعه نمونه یک هکتاری در هر یک از تیپ های مذکور جنگلی ناو اسالم گیلان که از لحاظ شرایط محیطی همگن بودند، آماربرداری شدند و با استفاده از نرم افزار (Crancod ver 1.4) شاخص های میانگین جهت، زاویه یکنواخت، آمیختگی، ابعاد قطر برابر سینه و ارتفاع و فاصله تا نزدیک ترین همسایه ها محاسبه شدند. نتایج حاصل از شاخص های میانگین جهت و زاویه یکنواخت، نشان دهنده موقعیت مکانی تصادفی برای درختان سه تیپ مورد بررسی بود. نتایج شاخص آمیختگی، اختلاط کمی را برای سه تیپ مورد بررسی نشان داد. نتایج به دست آمده از این شاخص نشان داد که غالبا گونه Fagus orientalis دارای رقابت درون گونه ای و سایر گونه های مورد بررسی دارای رقابت بین گونه ای می باشند. میانگین شاخصهای ابعاد قطر برابر سینه و ارتفاع، شباهت نسبی تیپهای مورد بررسی را نشان داد. نتایج حاصل از این دو شاخص غالبیت نسبی گونه های Acer velutinum و Alnus subcordata و مغلوبیت گونه های Cerasus avium و Ulmus glabra را نشان دادند. مقایسه ساختار گونه های مشابه در تیپهای مختلف، بیانگر عدم وجود اختلاف معنی دار بین آنها از نظر ویژگی های موقعیت مکانی، ابعاد قطر برابر سینه و ارتفاع و تراکم درختان بود

شناسایی برخی گونه های کاردامینه در ایران با استفاده از نشانگر ملکولی ITS
جنسCardamine L. از خانواده Brassicaceaeبا بیش از 200 گونه در جهان و 7 گونه در ایران با توجه به تنوع کاریولوژی و مورفولوژی بالا، گیاه مناسبی برای بررسی های فیلوژنتیکی و تکامل می باشد. گونه کاردامینه هیرسوتا از این جنس بتازگی بعنوان گیاهی مدل در مطالعات مقایسه ای تکاملی معرفی شده است. این گیاه از هفت منطقه مختلف ایران جمع آوری و پس از استخراج DNA ی برگی، ناحیه ITS آنها با استفاده از پرایمر های ITS4 وITS5 تکثیر و سپس تعیین توالی گردید. توالی ها با استفاده از نرم افزار مگا 7 مورد آنالیر فیلوژنتیک قرار گرفتند و از طریق مقایسه نتایج بلاست و درخت فیلوژنتیکی، گونه های C.hirsuta C.flexuosa, C.tenera,و C.uliginosa شناسایی گردیدند. همچنین با استفاده از دو اکوتیپ اروپایی مشخص از گونه هیرسوتا به عنوان شاهد (آزروس و اکسفورد تهیه شده از موسسه ماکس پلانک)، نتایج حاصل تایید گردید. یافته های این پژوهش، علاوه بر تایید حداقل چهار گونه کاردامینه در ایران، کارآمدی نشانگر ITS را در تفکیک گونه های مختلف جنس کاردامینه بخوبی نشان داد.

ریز ازدیادی گیاه دارویی آب بشقابی L.) (Centella asiatica
Centella asiatica (L.) از جمله گیاهان دارویی پرمصرف و محبوب در سراسر دنیا است که در خاورمیانه، اروپا و آمریکا دارای محبوبیت فراوانی است. این گیاه دارای خواص ضد باکتری و ضد قارچی بوده و در درمان آسم و برونشیت نیز به کار میرود. در سالهای اخیر به دلیل بهرهبرداری نامحدود و بدون برنامه از این منبع گیاهی، و تلاشهای ناکارآمد در احیای این گونه گیاهی دارویی ارزشمند، ذخیره گیاهی Centella asiatica (L.) در طبیعت به طور چشمگیری کاهش یافته و در معرض خطر نابودی قرار دارد. در همین راستا علاقه به تکنیکهای کشت in vitro از جمله تکنیک ریزازدیادی از طریق کشت بافت جهت تکثیر این گیاه ارزشمند افزایش یافته است. در این پژوهش نیز ریزازدیادی این گیاه از طریق کشت بافت گیاهی برای نخستین بار در ایران توسط ریزنمونههای گرهای بررسی شد. جهت تکثیر، بخشهای گرهای از نمونههای جمعآوری شده از عرصه در استان گیلان، پس از سترونسازی در محیط کشت MS حاوی -1 mgL1 BA و -1 mgL 1/0 IBA مستقر گردیدند. پس از یک ماه، ریزنمونههای رشد یافته به منظور دستیابی به بهترین تیمار شاخهزایی، تحت 9 تیمار هورمونی مختلف مورد آزمایش قرار گرفتند. با بررسی نتایج، محیط کشت MS حاوی -1 mgL5/0 BA و -1 mgL 5/0 2ip و -1 mgL 1/0 NAA به عنوان تیمار برتر شاخهزایی معرفی شد. جهت ریشه زایی و سازگاری در گلخانه، بررسی 11 تیمار هورمونی مختلف در محیط کشت MS نشان داد که تیمار حاوی -1 mgL5/0 IBA بهترین نتایج را در برداشته است.

تعیین شرایط بهینه برای ریزازدیادی گیاه زالزالک(Cratagus aronia) در شرایط کشت درون شیشهای
زالزالک از تیره گل سرخ (Rosaceae) و ازجمله گیاهانی است که دارای مصارف دارویی و زینتی بوده و ارزش صادراتی دارد. این جنس به دلیل انجام دورگه شدههای فراوان، دور گیری درهم، سیستم تولیدمثلی آپومیکسی و پلی پلوییدی به لحاظ تکثیر یکی از مشکلترین جنسها برای گیاهشناسان است. استفاده از فناوریهای نوین بخصوص تکنیک کشت بافت در تولیدات گیاهی قادر خواهد بود بهعنوان روش جایگزین، روند تولید انبوه و کنترلشده این گیاه را ارتقا بخشد. از این رو در این تحقیق مراحل مربوط به بهینه سازی ریز ازدیادی گونه Crataegus aronia شامل شاخه زایی و ریشهزایی در شرایط کشت درون شیشهای مورد ارزیابی قرارگرفته است. نمونهبرداری در فصل بهار انجام و سپس سترونسازی شدند. محیط کشت پایه (MS) جهت استقرار اولیه مورداستفاده قرار گرفت. آزمایش شاخهزایی در محیط کشت MS و غلظتهای مختلفی از تنظیمکنندههای رشد ایندول بوتیریک اسید، ایندول استیک اسید، بنزیل آمینو پورین و تیدیازورون و آزمایش ریشهزایی در محیطهای 2/1 MS، MS با غلظتهای مختلف ایندول بوتیریک اسید و نفتالین استیک اسید صورت گرفت. نتایج مقایسه میانگینها نشان داد که بهترین میزان شاخهزایی درتیمار 3 میلیگرم در لیتر بنزیل آمینو پورین و 1 میلیگرم در لیتر ایندول بوتیریک اسید و بیشترین میزان ریشهزایی در محیط کشت 2/1 MS در تیمار 3 میلیگرم در لیتر ایندول بوتیریک اسید حاصل شد.

بررسی اثر آهن، نوع محیط کشت و ترکیب هورمونی محیط کشت بر باززایی دو گونه رز Rosa canina و Rosa beggeriana در شرایط این ویترو
دو گونه Rosa canina و Rosa beggeriana به علت دارا بودن خصوصیات مطلوب از جمله رشد سریع و معطر بودن اهمیت قابل توجهی دارند. با توجه به مشکلات تکثیر رز از طریق روشهای سنتی، استفاده از روش های ریزازدیادی توصیه شده است. از جمله مشکلات کشت این ویترو نیز می توان به پدیده زردشدگی برگ ها اشاره نمود. لذا در این پژوهش جهت افزایش سبزینگی برگها، دو آزمایش طراحی شد. در آزمایش اول محیط کشت MS تغییر یافته (آهن به مقدار سه برابر (34/8 میلی گرم در لیتر) و 30 میلی گرم در لیتر نیترات نقره ) به همراه دو نوع هورمون BAP و TDZ مورد بررسی قرار گرفت. نتایج آزمایش اول نشان داد، بیشترین درصد سبزینگی برگها در محیط کشت حاوی 1 میلیگرم در لیتر هورمون BA و آهن سه برابر بود (6/42 درصد). طراحی آزمایش دوم بر روی نوع آهن (Fe-EDDHA و FeSO4) موجود در محیط MS به همراه ترکیب هورمونی 1 میلیگرم در لیتر BA و 5/0 میلیگرم در لیتر NAA انجام شد. نتایج نشان داد استفاده ترکیبی از ( Fe-EDDHA و FeSO4) توانسته بیشترین درصد سبزینگی برگها (15/67 درصد)، طول شاخه (92/3 سانتیمتر) و وزن خشک (03/0گرم) را تولید نماید. گیاهچههای باززایی شده به منظور ریشهزایی به محیط کشتهای MS2/1جامد و مایع حاوی 40 گرم در لیتر ساکارز، 25/0 میلیگرم در لیتر هورمون BA، 1 میلیگرم در لیتر هورمون IBA و 1/0 میلیگرم در لیتر هورمون NAA انتقال یافتند. بیشترین تعداد ریشه تولید شده (1/9) و طول ریشه (4/3 سانتیمتر) در محیط کشت جامد مشاهده شد.

شمارش کروموزومی چند گونه از نهاندانگان ایران
10 گونه با 12 جمعیت از خانوادههای مختلف بومی ایران مورد بررسی قرار گرفتند. بررسب عدد کروموزومی گیاهان کمک شایانی در بدست آوردن اطلاعات تکاملی آنها می نماید.هدف از این بررسی بدست آوردن عدد کروموزومی گونهها و جمعیتهای مختلف گیاهان بود. بذر گونه ها از مناطق مختلف ایران جمع آوری گردید. دو گونه Melica persica و Silene vulgaris دارای دو جمعیت است. عدد کروموزومی Papaver rhoeas (14=n2) و Filipendula vulgaris (28=n2) برای اولین بار از ایران گزارش شدند. چهار گونه Silene vulgaris (24=n2)، Phlomis olivieri (20=n2)، Vicia armena (10=n2) و Melica persica (18=n2) برای دومین بار از ایران گزارش شدند. Tanacetum pinnatum (18=n2)، Vicia persica (10=n2)، Salvia hypoleuca (22=n2 ) و Papaver bracteatum (14=n2 ) دیگر گونههایی بودند، که عدد کروموزومی آنها مورد بررسی قرار گرفت. تجزیه و تحلیل داده ها با استفاده از نرم افزار MicroMeasure و Excel صورت گرفت. ایدیوگرام برای همه جمعیتها رسم گردید. فرمول کاریوتیپی همه گونه ها ارائه شد.

رابطه ویژگیهای خاک با قارچهای میکوریز آربسکولار همزیست با زالزالک (Crataegus pontica)
همزیستی قارچهای میکوریز با ریشهی گیاهان عالی، یکی از مهمترین انواع همزیستی اجباری در طبیعت بوده که فواید بسیاری از جمله جذب آب و عناصر غذایی برای گیاه میزبان دارد. هدف از این مطالعه، بررسی رابطهی فراوانی اسپورها و کلینیزاسیون ریشهی درخت زالزالک با عوامل محیطی در جنگلهای زاگرس است. بدین منظور در مناطق دینارکوه در شهرستان آبدانان و ارغوان در شهرستان ایلام، 54 نمونه از خاک به همراه ریشه از عمق 30-0 سانتیمتری تهیه گردید. نمونههای خاک جهت استخراج و شناسایی قارچهای همزیست، اندازهگیری فراوانی اسپور قارچها، تعیین درصد کلینیزاسیون و اندازهگیری ویژگیهای فیزیکی و شیمیایی خاک (بافت خاک، وزن مخصوص ظاهری، ضخامت لاشبرگ، اسیدیته، شوری، پتاسیم قابل جذب، فسفر ،ماده آلی، نیتروژن) به آزمایشگاه منتقل شدند. نتایج نشان داد که در مجموع 13 گونه قارچ میکوریز با درختان زالزالک همزیستی دارند که گونههای Claroideoglomus etunicatum و Glomus caesaris بیشترین فراوانی و گونههای Acaulospora thomii و Entrophospora infrequens کمترین فراوانی را دارا بودند. نتایج همچنین نشان داد که فراوانی اسپور قارچ ها در رویشگاه دینارکوه از رویشگاه ارغوان بیشتر میباشد. نتایج همبستگی پیرسون نشان داد که بین درصد کلینیزاسیون و فراونی اسپور با پتاسیم و pH خاک همبستگی مثبت معنیداری وجود دارد. نتایج آنالیز تحلیل ﻣﺆلفههای اصلی نیز نشان داد که درصد کلینیزاسیون و فراونی اسپور قارچها از بین عوامل محیطی تنها با پتاسیم و میزان لاشبرگ همبستگی معنیدار مثبتی دارد.
