Stevia (Stevia rebaudiana) is a sweet medicinal herb that is cultivated in many parts of world and may experiences low temperature in winter. In this study, we aimed to identify physiological and biochemical changes that occur in Stevia exposed to low temperature and enable the plants to survive at low temperature. Stevia grown in vitro and incubated under controlled conditions at 5, 10 and 25 ºC for one month and then physiological and biochemical indicators in leave and shoot were measured. Low temperature decreased the contents of chlorophyll a, chlorophyll b, total chlorophyll and carotenoids in the leaves. In addition, we observed increased accumulation of hydrogen peroxide and malondialdehyde in leaves and shoot of Stevia exposed to low temperature. Low temperature increased the activities of Superoxide dismutase, catalase, ascorbate peroxidase, polyphenol oxidase and guaiacol peroxidase in leaves and shoot of Stevia. Low temperature lead to increased accumulation of secondary metabolites (phenoloc compounds, flavonoids and anthocyanins) and also increased activities of the enzymes of their biosynthesis pathway (phenylalanine ammonia lyase and Tyrosine ammonia lyase) in leaves and shot of Stevia plants. In response to low temperature, Stevia plant increased the accumulation of total soluble sugar, reducing sugar, total amino acids and proline in their leaves and shoot. In general, to cope with low temperature, Stevia evolved a variety of physiological and biochemical mechanisms that can be suggested to use for identifying cold-tolerant stevia cultivars.}, keywords = {Low temperature,Stevia,oxidative Stress,Antioxidant system,Reactive oxygen species}, title_fa = {بررسی تغییرات فیزیولوژیکی و بیوشیمیایی در برگ و ساقه گیاه استویا تحت تنش دمای پایین}, abstract_fa = {دمای پایین یکی از فاکتورهای مهم محدود کننده رشد، نمو و پراکنش گیاهان است. استویا (Stevia rebaudiana) یک گیاه علفی دارویی شیرین است که در بسیاری از مناطق جهان کشت می‌شود و ممکن است تنش دمای پایین را در زمستان تجربه کند. در این مطالعه، تغییرات فیزیولوژیکی و بیوشیمیایی که در گیاهان استویا تحت تنش دمای پایین رخ می‌دهد و باعث بقاء آنها می‌شود مورد بررسی قرار گرفت. استویا برای یک ماه تحت تیمارهای دمایی 5، 10 و 25 درجه سانتیگراد قرار داده شد و شاخص‌های فیزیولوژیکی و بیوشیمیایی در برگ و ساقه اندازه‌گیری شد. در دمای پایین محتوی کلروفیل a، کلروفیل b، کلروفیل کل و کاروتنوئیدها در برگ کاهش پیدا کرد. به علاوه، تجمع افزایش یافته پراکسید هیدروژن و مالون دآلدئید در برگ و ساقه استویا در معرض تنش دمای پایین مشاهده شد. تنش دمای پایین فعالیت آنزیم‌های سوپراکسید دیسموتاز، کاتالاز، آسکوربات پراکسیداز، گایاکول پراکسیداز و پلی‌فنل اکسیداز را در برگ و ساقه استویا افزایش داد. تنش دمای پایین به تجمع متابولیت‌های ثانویه (ترکیبات فنلی، فلاونوئیدها و آنتوسیانین‌ها) و فعالیت افزایش یافته آنزیم‌های کلید مسیر بیوسنتزشان (پلی‌فنیل آمونیالیاز و تیروزین آمونیالیاز) در برگ و ساقه استویا منجر شد. در پاسخ به تنش دمای پایین، استویا تجمع قند محلول کل، قند احیاء آمینواسید کل و پرولین را در برگ و ساقه افزایش دادند. به طور کلی، برای غلبه بر آسیب‌های ناشی از تنش دمای پایین، مجموعه‌ای از مکانیسم‌های فیزیولوژیکی و بیوشیمیایی در گیاه استویا فعال شد که این مکانیسم‌ها می‌توانند برای شناسایی رقم‌های استویا مقاوم به دمای پایین استفاده شوند.}, keywords_fa = {دمای پایین,استویا,تنش اکسیداتیو,سیستم آنتی‌اکسیدانت,گونه‌ها فعال اکسیژن}, url = {https://plant.ijbio.ir/article_1689.html}, eprint = {https://plant.ijbio.ir/article_1689_094518ff34f497e8c2ac46ffc96bd944.pdf} }
