Legume crops are of great significance because they can use nitrogen from the atmosphere through biological nitrogen fixation. In the current study, effects of NARK gene and different nitrogen treatments on growth, physiological parameters and protein profile of soybean leaf of supernodulation mutant and wild-type were investigated. For this, a factorial experiment in a completely randomized design with three replications was conducted under greenhouse conditions. Experiment factors included two levels of ammonium (NH4+) and two levels of nitrate (NO3-) for three types of soybean (sterile, wild-type and supernodulating mutant). Results of variance analysis revealed that the treatment of ammonium plus nitrate in comparison with other treatments had the greatest effect on soybean plants in terms of morphological and biochemical parameters. Two-dimensional electrophoresis also showed that sterile plants under 5mM ammonium sulfate treatment had higher protein variation in leaf compared with the soybean wild-type plants inoculated with rhizobia (non-recipient of mineral nitrogen). This variation in the leaf protein profile of plants could be due to type of nitrogen they receive (mineral nitrogen/biological nitrogen fixation). In addition, the protein variation could be due to alteration in genes expression because of symbiosis in inoculated plants with rhizobia. Traits such as chlorophyll b, total chlorophyll, carotenoids, and reduced sugar content, had the highest amount under the ammonium plus nitrate and 5mM ammonium treatments and the lowest amount in control plants.}, keywords = {Biological nitrogen fixation,NARK,Soybean,protein profile,nodulation}, title_fa = {تأثیر ژن NARK و تیمار نیتروژن بر رشد، و پروفایل پروتئینی برگ در تیپ موتانت سوپر گره‌زا و تیپ وحشی سویا}, abstract_fa = {نیتروژن یکی از مهم ترین عناصر مورد نیاز گیاهان می‌باشد. گیاهان لگوم بدلیل دریافت این عنصر از طریق تثبیت بیولوژیک نیتروژن اتمسفر حائز اهمیت فراوان می‌باشند. در این مطالعه، تأثیر ژن NARK و تیمارهای نیتروژن‌دار بر رشد و پارامترهای فیزیولوژیک و همچنین تغییرات پروتئینی برگ گیاه سویا تیپ وحشی با تیپ موتانت بررسی گردید. بدین منظور، آزمایش گلخانه ای بصورت فاکتوریل و در قالب طرح کاملاً تصادفی با سه تکرار به اجرا در آمد. تیمارهای آزمایش شامل دو سطح آمونیوم (NH4+) و دو سطح نیترات (NO3-) برای سه تیپ گیاه سویا، استریل (عدم تلقیح با باکتری ریزوبیوم)، وحشی و موتانت سوپر گره‌زا بودند. نتایج تجزیه واریانس نشان داد که تیمار آمونیوم به همراه نیترات نسبت به سایر تیمارها بیشترین تأثیر را بر گیاهان سویا از نظر شاخص های رشد و صفات بیوشیمیایی گذاشته است. الکتروفورز دو بُعدی نیز نشان داد که در گیاهان تیپ استریل تحت تیمار 5 میلی‌مولار سولفات آمونیوم تنوع پروتئینی بیشتری در برگ نسبت به گیاهان تیپ وحشی تلقیح شده با باکتری ریزوبیوم (عدم دریافت نیتروژن معدنی) وجود دارد. این تنوع در پروفایل پروتئینی برگ گیاهان می‌تواند بدلیل نوع نیتروژن دریافتی گیاهان (نیتروژن معدنی/تثبیت بیولوژیک ازت) باشد. علاوه بر این، تنوع پروتئینی می تواند بدلیل تغییر در بیان ژن‌ها بدلیل همزیستی گیاهان تلقیح شده با باکتری ریزوبیوم باشد. سایر صفات مورد بررسی تحت تیمارها از قبیل کلروفیل b، کلروفیل کل، کارتنوئید، قند‌های احیا‌کننده، بیشترین مقدار را در تیمار توأم و تیمار 5 میلی‌مولار سولفات آمونیوم و کمترین مقدار خود را در تیمار شاهد نشان دادند.}, keywords_fa = {تثبیت بیولوژیک ازت,NARK,سویا,پروفایل پروتئینی,گره‌زایی}, url = {https://plant.ijbio.ir/article_1554.html}, eprint = {https://plant.ijbio.ir/article_1554_3fc1625f79871520aaec6160ea2214b8.pdf} }
